It takes the match-winning streak up to eight for the Chilean, who is on a hugely impressive run of form having taken the title in Geneva the week prior to Roland-Garros.
Jarry, ranked No 35, will face Norwegian Casper Ruud, the No 4 seed, next.
Ahead of his victory, the Chilean beat Hugo Dellien (6-4, 6-4, 6-2) and edged out American Tommy Paul, the No 16 seed (3-6, 6-1, 6-4, 7-5).
Giron, ranked No 75, won against Serb qualifier Hamad Medjedovic (6-0, 6-2, 1-6, 6-0) and Czech Jiri Lehecka (6-2, 6-3, 6-2) earlier in the tournament.
